There are several ways to get more Dwellers in Fallout Shelter, but some are more reliable than others.
1. Dwellers Will Arrive Over Time
You'll get more Dwellers over time as it passes; more Dwellers will find their way from the Wasteland into your Vault. However, there are much quicker ways to get more Dwellers in Fallout Shelter.
2. Make Vault Dweller Babies
The best and most optimal way to get more Dwellers is to breed them. Female Dwellers will be pregnant for three hours, at which point they'll give birth, and it'll take another three hours for the child to grow up and be able to perform activities in your Vault.
3. Unlock Dwellers As Special Rewards
You can also get Dwellers as rewards for completing various tasks, such as quests, and as Lunchbox rewards. While this isn't quite as reliable a way to get more Dwellers as breeding them, it's still a good way to get a few more Dwellers in your Vault.
As you play Fallout Shelter, you'll inevitably get more Dwellers over time. However, making the right moves in the game can help you get more Dwellers much faster, allowing you to get way more resources and reap the benefits of having a huge Vault.
